I would recommend turning it off the first day after installation.
I can hardly tell there is a protective film on my surface, and it doesn't affect the pressure sensitivity of the pen. If you are using a snug fitting case I would recommend taking it off the first day after installing the screen protector as the corners will raise if it is not perfectly centred.
